چکیده

Liver cancer is one of the most common solid tumors worldwide. Extensive research was carried out to document the powerful properties of Nigella sativa (N. sativa) as an anti-tumor, bactericide, anticestode, antinematode, anti-inflammatory, analgesic, anti-diabetic and diuretic with many other uses. Little is known about the Nigella sativa anti-tumor property in liver cancers, thus our current study was performed to investigate the protective role of Nigella sativa in DAB induced liver carcinogenesis. 140 male Albino mice weighing 40-50 gm divided into four groups. Group one was normal control group without treatment of any type. Group two was Nigella sativa treated control group. Group three was treated with DAB carcinogen. Group four was treated with both Nigella sativa and DAB. Biochemical investigations, flow cytometric analysis, and histopathological examination of the liver tissue were performed for all groups. The results showed that there was a significant change in the DNA content, histomorphology, and antioxidant enzymes in the liver tissues of the DAB treated group. These changes were restored to approximately the normal counterpart with Nigella sativa treatment. In addition, treatment with Nigella sativa only showed comparable results with control untreated groups on different levels. Collectively, these results give clear evidence that Nigella sativa lonely induce no harmful effects on the liver. Moreover, it exerts hepatoprotective effect against liver carcinogens. Antioxidant property is mediated its actions and investigating other underlying mechanisms merits further studies. [Nature and Science 2010;8(6):80-87]. (ISSN: 1545-0740).

Spectrophotometric analysis of cytochromes in rat liver during carcinogenesis.

The cytochrome content of liver mitochondria and microsomes during the carcinogenesis of rat liver by both 3'-methyl-4-dimethylaminoazobenzene (3'-DAB) and 4'methyl-4-dimethylaminoazobenzene (4'-DAB), and of 3'DAB-induced hepatomas, was determined spectrophotometrically at room and liquid nitrogen temperatures and was compared with that of normal liver. The cyto chrome a + a3 content in the liv...

Effect of Thymoquinone and Nigella sativa Seeds Oil on Ethanol Toxicity in Rats

Background: In this study, the protective effect of thymoquinone (TQ) and Nigella sativa seeds oil (NSO) was evaluated against oxidative damages induced by ethanol in rats. Methods: Animals were treated with ethanol 40% daily by gavages once a day for 4 weeks. NSO and TQ were injected intraperitoneal once a day for 4 weeks. The histopathological examination of liver, kidney, brain and heart was...

To study the influence of activation of metabolic function of carcinogen to chemical carcinogenesis, effect of barbital, an inducer of enzymes related to drug metabolism, was studied on the development of liver cancer in the rat fed with 4-dimethylaminoazobenzene (DAB), and compared with that of 3-methylcholanthrene,*4 known to antagonize the carcinogenic effect of DAB. In an animal experiment ...

Analysis of loss of nuclear RNA in azo dye-induced hepatoma by DNA-RNA competitive hybridization.

DNA-RNA hybridization studies, using nuclear RNA's (nRNA's) labeled in vivo and in vitro with high specific radioactivities, were performed to compare the nRNA populations of normal rat liver, livers treated with 3'-methyl-4-dimethylaminoazobenzene (3'-Me-DAB), and 3'-Me-DAB-induced hepatomas. The study with normal liver nRNA labeled by i.p. injection of [3H]orotic acid indicated that the nucle...

Carcinogenesis in tissue culture. 22. Malignant transformation of cloned rat liver cells treated in culture with 4-dimethylaminoazobenzene and properties of the transformed cells.

Cultured rat liver cells which were cloned from a single cell were transformed into malignant cells by a chemical carcinogen, 4-dimethylaminoazobenzene (DAB). The DAB-transformed cells produced tumors when back-transplanted into new-born rats but the carcinogen-untreated control cells did not. Characteristics of the transformed liver cells were compared to those of DABuntreated control cells in...
